Realized that the medallion Discord gave her is the last key needed to open the Chest of Harmony. Ordered Lord Tirek to release her friends including Discord in exchange to giving him her Alicorn magic from the other princesses. Fought Lord Tirek when he destroyed the Golden Oaks Library. Kept Princess Celestia, Princess Luna, and Princess Cadance's Alicorn magic hidden from Lord Tirek by having the Alicorn magic be given to her. Drank Zecora's potion to find out as to why Princess Celestia and Princess Luna disappeared.

While Queen Chrysalis is distracted proclaiming her victory of taking over Canterlot, Twilight Sparkle took the time to free Cadance and have Cadance break Shining Armor's spell from Queen Chrysalis. Fought the Changelings alongside her friends in an attempt to get the Elements of Harmony. Used a bouquet of flowers to distract Minuette, Lyra Heartstrings, and Twinkleshine from getting in her and Cadance's way. Teamed up with Cadance to escape the caves and and stop the imposter Cadance from marrying Shining Armor. Found out that the real Cadance is with her in the caves and that the other Cadance is an imposter. After being sent down to the caves beneath Canterlot by Cadance, Twilight Sparkle shoots magic towards the Cadance appearing in the crystal reflections. Found out that Cadance is brainwashing Shining Armor. Suspected that there was something wrong with Cadance's behavior towards her, her friends, and Shining Armor. She and her friends used the Elements of Harmony to defeat Discord. Freed her friends from Discord's corruption. Regained her hope on friendship when Princess Celestia sent her the friendship lessons she had given to Princess Celestia. Attempted to have her friends use the Elements of Harmony despite them being in their corrupted state to defeat Discord. Realized that the Elements of Harmony are in her Golden Oak Library. She and her friends accepted Discord's challenge on find the Elements of Harmony. Uses the Elements of Harmony to destroy Nightmare Moon and have Princess Luna redeem herself. Learned the magic of friendship and learned that the sixth Element of the Elements of Harmory is magic.
